Rheonix uniquely incorporates a "Hy-Fi" microarray panel approach to complex diagnostic situations. The traditional process tests a patient specimen for a single disease causing organism, such as FLU. A negative result generates further tests. With the Rheonix panel, patients can be tested for an array of diseases simultaneously yielding a quick diagnosis and rapid treatment. The Rheonix panel is driven by a micro-spotted array of capture probes embedded in the Rheonix CARD. Up to 48 probes can be used to develop a powerful panel of information. Here's how the microarray works: 1. The first step is to denature the biotinylated amplicons, which result from PCR amplification. This is accomplished through the addition of precisely controlled temperature. 2. The denatured amplicons are passed over the capture probes where complimentary bonding between base pairs of the probe and amplicon result in target capture. 3. Finally, the bonded pairs are washed to remove excess amplicons and any waste material. 4. The streptavidin, conjugated with horseradish peroxidase, attaches to the biotin. This arms the complex preparing it for detection. 5. Excess is removed. The Streptavidin Horseradish Peroxidase conjugated probe and amplicon pairs remain for visualization. 6. The final step adds tetra methyl benzidine, a chromogenic substrate, which when activated by horseradish peroxidase, forms a blue spot on the microarray indicating target capture. In this example, the TMB reveals the presence of Rhinovirus.
