Enregistrée entre décembre 1966 et janvier 1967, c’est la deuxième chanson — après Strawberry Fields Forever — que les Beatles enregistrent pour leur prochain album, qui ne porte pas encore de nom mais qui deviendra Sgt. Pepper’s Lonely Hearts Club Band. Toutefois, les deux titres n’y figureront pas, ayant été précédemment publiés ensemble en 45 tours « double face A » ; leur manager Brian Epstein voulait en effet maintenir la popularité du groupe après une inhabituelle période de six mois sans publication inédite. Le single sort ainsi le 13 février 1967 aux États-Unis et le 17 février au Royaume-Uni. Au mois de novembre 1967, Penny Lane est à nouveau publiée sur la version américaine de l’album Magical Mystery Tour. Penny Lane et Strawberry Fields partagent le thème de la nostalgie de l’enfance de leurs deux auteurs dans leur ville natale de Liverpool. Le 18 mars 1967, le disque est numéro un des ventes aux États-Unis, mais — et c’est une première depuis quatre ans — n’atteint que la seconde place des hit-parades britanniques.
