Welcome to the LIBREhub Seminars “OpenScopes: an open, modular platform to widen access in advanced microscopy" By Paul French Professor of Physics Imperial College London Overview In this video, we review recent developments and applications of multidimensional fluorescence and phase imaging instrumentation, and outline ongoing efforts to widen access to these advanced imaging modalities, including high-content analysis (HCA), through the “openScopes” open instrumentation platform (www.openscopes.com). openScopes includes research-grade instruments for multidimensional fluorescence and quantitative phase imaging, super-resolved microscopy, and optical projection tomography. To further broaden access, an open-source modular microscope stand, “openFrame,” has been developed. This platform can be used flexibly to create affordable and sustainable instruments for lower-resource settings (e.g., LMICs) or for rapid prototyping of advanced microscopy concepts. A basic openFrame-based brightfield/fluorescence microscope can be upgraded to support almost any microscopy modality. For HCA and slide scanning applications (e.g., histopathology), a novel open-source optical autofocus module has been developed. Advanced openFrame-based instruments include automated single-molecule localization microscopy (SMLM) based on easySTORM, as well as a modular multiphoton multiwell plate microscope (M³M). This system incorporates multiplexed TCSPC-FLIM and quantitative phase imaging using a single-shot polarization differential phase contrast (pDPC) technique, enabling label-free single-cell segmentation and tracking during long-term time-lapse fluorescence (FRET) assays. --------------------------------------------------------------------------------------------------------------------- LIBREhub seminar series bring and connect international speakers with the Latin American bioimaging community through a wide range of topics related to bioimaging technologies.
