Single Molecule Techniques Speaker: Chris Johnson, MRC Laboratory of Molecular Biology, UK The LMB Biophysics Facility houses a wide range of state-of-the-art and in-house built instruments that enable the molecular characterisation of macromolecular size, shape, mass, stability, binding affinity, dynamics and interaction kinetics. The facility is primarily intended for hands-on research by students, postdocs, and group leaders, who prepare samples and make measurements. Biophysics supports users in advice on experimental techniques and results, training and in collaborative research. In this video Chris explains why single molecule measurement should be untaken, the practicalities involved and several applications using the current single-molecule set-ups in the LMB including FRET and fluctuation correlation spectroscopy (FCS). He also details a new single molecule light scattering technique using the Refeyn iSCAT to measure masses. In the final part of the video, he introduces a recently installed instrument, the LUMICKS C-Trap, which combines optical tweezers and laminal flow fluidics to manipulate and study forces between biomolecules.
