The short answer...round core strings are generally more flexible, with warmer vintage tones and a longer sustain. Hex core strings are slightly stiffer and generally have a brighter attack with very consistent tone, but shorter sustain. Round cores have 100% contact between the core wire and wrap wire making them more dense. Great for sustain. All strings used to be made this way which is why their sound is considered vintage by some. The round core is more flexible due to lower tension and freedom for the wrap wire to move with the core. Because of this, players should always tune to pitch round core strings before trimming ends. Round core strings can slip if cut before tuning, leaving you with a useless expensive string. Once hex cores were introduced, they produced such consistent tone due to the precise process of the wrap wire holding tightly to the angular core, and then maintaining that tension, that they quickly became the industry standard. Like all strings, it really comes down to what you like and the sound you are looking for.
