In this video Elias gives you a detailed, step-by-step guide on how to perform the DNS glucose assay. Please refer to his written protocol for a more concise guide. He was unable to develop color when he performed the assay this time around. After some trouble shooting, he was able to determine that this was due to not enough base (i.e., NaOH) being added to the DNS solution. The ultimate source of this error was an uncalibrated balance. At any rate, assuming that you add 1 g of NaOH pellets as you're supposed to (and that your balance is accurate), you should be able to develop color. As a reminder, once you have developed color in your standards and samples you need to analyze them using a spectrophotometer.
