Pharmacologists classify buspirone as a serotonin 1A partial agonist, as its primary action appears to be the activation of pre-synaptic serotonin receptors, which gradually decreases firing of serotonin neurons. For years, buspirone has been considered a third or fourth tier option for treating ADHD. In addition to effectively treating anxiety associated with ADHD, it may also provide direct benefit for ADHD. 1-2-pyrimidinylpiperazine (1-PP), a major metabolite of buspirone, is an alpha-2 antagonist, which boosts levels of norepinephrine, dopamine and serotonin. Limited research indicates buspirone does reduce ADHD symptoms in humans. A handful of studies, almost all in rodents, indicate that buspirone can successfully treat several side effects associated with Ritalin, including teeth grinding (bruxism), liver toxicity, and growth inhibition. Buspirone also appears to reduce the potential for substance abuse and cognitive dulling that are associated with long term Ritalin use in rodents. In September 2023, the FDA approved gepirone, (Exxua) which is chemically similar to buspirone, for the treatment of depression.
