Northern blotting technique step by step - This lecture explains about the northern blotting procedure and technique to identify specific RNA fragment from a mixture of cellular RNA molecules separated by the formaldehyde treated Agarose gel electrophoresis. Northern blotting step by step explanation is provided to understand the procedure of the RNA separation and identification. The difference between Southern blotting and northern blotting is also explained in this video. Northern blotting consists of several different stages - 1. First is the isolation of mRNA from the target cell. 3. Then the separation of the native RNA molecules using formaldehyde treated Agarose gel electrophoresis so that all the RNA gets linearity. 4. Transfer of the Agarose gel RNA bands to a nitrocellulose or nylon Membrane filter. This filter paper after the RNA transfer is known as northern blot. 5. Then the use of complementary DNA or cDNA probe to bind with the target RNA that will indicate the presence of target sequence by radioactivity or by colorimetric analysis. Northern blotting was invented in 1979 at it was named after the southern blotting technology to identify RNA using blotting technique.
