Further evidence of IgG4 antibodies formation in mRNA vaccinated with new unwelcome twists introduced to the story. The study shows that boosting with mRNA vaccines appears desirable because, within a month of boosting, resulting antibodies were able to neutralize more viruses and against more variants, including the latest variants. Both of these appear very desirable outcomes of mRNA shots. However, two more aspects are also observed. It appears that all participants produced more IgG4 post mRNA XBB1.15 booster, including those that were infected prior to vaccination. Previously data suggested that such infection prior to vaccination offered protection from IgG4 antibody development. The study also looked at memory B cells targets and showed that mRNA boosting increased antibodies targeting other areas of the spike protein away from the receptor binding domain of the spike protein. This demonstrates that immune refocusing is taking place post mRNA boosting, meaning, the immune system is trained to target other areas of the spike protein more frequently than before. This is also 5th IgG4 study analyzed showing increased neutralization of more variants after mRNA boosting along with an increase in IgG4 antibody levels. This points to a potential autoimmune risk if the new areas of spike protein that are being targeted are similar to human proteins.
