We should always use HPLC grade or chromatographic grade solvents for the preparation of mobile phase. Always Use HPLC grade chloroform because HPLC grade chloroform does ethanol When we are using buffer solutions then we should make sure that buffer solution is clear, homogenous and free from particulate matter. While preparing buffer solutions proper care should be taken to select the correct reagent i.e anhydrous,monohydrous,dehydrate etc.If specification does not tells about hydrarte status then use monohydrous salt Adjust the Ph of the buffer solutions as per respective sop within +- 0.05 unless otherwise stated in SOP.And PH of buffer solution should always check after thorough mixing. Buffer solutions must and should be filtered through 0.45 micron filter or as mentioned in your test method. Friends if you will not use filtered buffer solution it will deposit your HPLC system tubings and you will phase lots of problem with respect to high pressure ghost peak etc. Buffer solutions should not be stored for longer duration they should always be prepared freshly. If you will store buffer solutions for longer duration then there will be bacteria and fungus growth Mobile phase should be degassed before usage but make sure that mobile phase is not kept under suction for longer duration otherwise highly volatile solvents will evaporate. Friends we should always use glass bottle for mobile phase preparation. We should always cover mobile phase container with suitable lid to avoid evaporation of the solvents. Mobile phase bottle shall be labelled legibly with details like date of preparation,valid up to date and compostion Do not expose any of your solvent bottle to sunlight. Always keep mobile phase bottle above solvent delivery system. Always use suction filters at the beginning of mobile phase tubing’s. Suction filters will prevent entry of particulate matter into your HPLC system. And make sure cleaning of the suction filters regularly by sonicating them with IPA and 1N Nitric acid. Friends during the changeover of the mobile phase on your hplc system please do not keep tubing’s directly into another mobile phase.
