Crawling of effector CD8+ T (CD8 TE) cells visualized by multiphoton intravital microscopy in the liver of a HBcAg transgenic mouse (H2b) that was injected with H2b-restricted Cor93 (red) and H2d-restricted Env28 (green) CD8 TE. Sinusoids are in gray and hepatocellular nuclei are in blue. Scale bar represents 50 mm. Observations indicate that CD8 TE exhibit an intrasinusoidal crawling behavior that halts when hepatocellular antigens are recognized. This is particularly relevant for the pathogenesis of viral infections such as those caused by HBV and HCV. Light sheet microscopy allows for fast high-resolution imaging of 3D biological samples, like whole rodent organs, embryos, or larvae. The use of paired orthogonal optical pathways to confine illumination to a single plane allows researchers to image sample volumes faster while limiting light dosage. This technique opens new avenues to study entire biological systems and processes in physiologically representative 3D samples.
