The use of rheological measurements to predict mucoadhesion is well established. In addition to characterising physical properties related to spreading and wetting, we can look at rheological changes brought about as mucoadhesive polymers intermingle with mucin proteins. This synergistic interactions create semi-permanent bonds that then promote the adhesion and cohesion necessary for sustained delivery of active pharmaceutical ingredients. The interaction and adhesion of a product to mucosa is a key requirement for the efficacy of many pharmaceutical and consumer health products, from ophthalmics to nasal and buccal gels, vaginal and gastrointestinal formulations. Rheological techniques deliver a powerful tool for researchers and product developers looking to demonstrate generic equivalence, support marketing claims or select mucoadhesive polymers to thicken a formulation. Oscillatory rheometry is one such applicable technique. Oscillatory testing entails applying small periodic stresses to a sample and observing its response. By performing measurements on the product of interest, along with a prepared mucin solution and a mix of the two, we can quantify the magnitude of the elastic response, and compare results for an unexpected difference indicating the degree of rheological synergism that promotes mucoadhesion.
