[TALK 19] Introduction to Mass Spectrometry - Holger Kramer - Biophysical Techniques Course 2022

Introduction to Mass Spectrometry Speaker: Holger Kramer, MRC Laboratory of Molecular Biology, UK The LMB Mass Spectrometry and Proteomics Facility houses a wide range of mass spectrometers enabling biological proteomics and structural proteomics. Biological proteomics involves the identification of proteins and their interactions and the comparative proteomics of cells and tissues. In structural proteomics purified proteins or their complexes are investigated by hydrogen-deuterium exchange or cross-linking mass spectrometry. In this video Holger begins with an overview of how samples are ionized, mass is analysed and detected by the mass spectrometer. He introduces fundamental concepts in mass spectrometry such as scan speed, transmission, mass accuracy and resolution. He describes the different types of instruments housed in the facility; how they differ and gives examples of applications such as protein identification, post-translation modification determination, structural proteomics and peptide mass identification. He then follows with a detailed description of the how different instrument ionise samples including: electron ionisation (EI), electrospray ionisation (ESI) and matrix-assisted laser desorption (MALDI). Finally, he shows how peptide fragmentation pattern is used in identification using the MASCOT database. The Mass Spectrometry and Proteomics Facility is headed by Holger Kramer.
