Gist: Here researcher wanted to optimized in-vivo CRISPR knockout on tumor restricted transgene expression. They started by designing target gene downstream of the pdcd1 start codon, which is expressed more in the activated T cell. Initially they checked with GFP expression in the activated T cell to check its functionality. We can see increased GFP expression in the activated T cells (panel c and panel d). Once validating the in-vitro results, then checked their system in-vivo to check the specificity, in which check the GFP expression level in the spleen and near tumor, and found activation and expression levels are high near tumor (panel f and e). After validating the specificity in-vivo with GFP, they use their system to form multiple cytokines to kill tumors such as IFN-Y, IL-2,IL-12 etc. Although, they see high expression of these cytokines in tumor specific area, In the case of IL-12 they found side effect in terms of weight loss, which they attribute to high peripheral transgene expression and need to change the promoter pdcd1. #crispr #invivo #cancer #cancertreatment #cancerawareness #pdcd1gene #biology #tumor #sideeffects #transgeneexpression #GFPexpression #tumorrestrictedpromoter Note: This is not a in-vivo CRISPR KO, rather in-vitro CRISPR KO.
