Description of the video: In this video I walk you through the process of making fluorescent microscopic images. I take a fluorescent brain sample in which I previously stained two set of cells. One set is labelled with a neuronal tracer while the other set naturally contains the protein calretinin. However, neither the tracer nor calretinin is visible in itself – we must stain them with a technique called fluorescent immunohistochemistry. We use special immunoglobulins fused with colorful molecules that specifically bind to the tracer or to calretinin. When we shine different color light beams on these samples these colorful molecules start to glow – but only one at a time depending on the color of the light we shine on them. So, we can capture a channel in which only the tracer and another one in which only the calretinin molecules can be seen. Finally, these channels are merged into one fluorescent image.
