National Institute on Aging, NIH (USA) Introduction by Steven Kleinstein (Yale University) The primary adaptive immune repertoire is generated during lymphocyte development via recombination of gene segments that are widely separated in the genome. For immunoglobulin heavy chain (IgH) genes two DNA recombination events bring together a variable (VH), a diversity (DH) and a joining (JH) gene segment to encode the variable domain of antibody heavy chains. Quasi-random assortment of multiple germline encoded VH, DH and JH gene segments during VDJ recombination provides the first level of antibody diversity. Optimal use of these gene segments for generating a diverse repertoire is imposed by genetic and epigenetic mechanisms. The role of chromatin configuration mediated by cis-regulatory sequences and trans-acting factors that ensure a diverse IgH repertoire will be discussed.
