This video demonstrates the loading sequence for running immobilized pH gradient (IPG) strips in the PROTEAN i12 system for isoelectric focusing (IEF) in the first dimension of two-dimensional (2-D) protein electrophoresis. The quick, easy, and foolproof loading and assembly of the focusing tray is illustrated step by step for the gel-side-down configuration; disassembly is also shown. The PROTEAN i12 IEF system can run up to 12 IPG strips from 7--24 cm long. Each channel in the i12 focusing tray is independently powered, giving the user total control over each IPG strip.
