EcoRI is a type II restriction endonuclease isolated from the bacterium Escherichia coli strain RY13. This enzyme plays a crucial role in molecular biology and genetic engineering due to its ability to cleave DNA at specific recognition sites. EcoRI recognizes and cuts the DNA sequence 5'-G↓AATTC-3', producing "sticky ends" with 5' overhangs of AATT. Structurally, it is a homodimer consisting of two 31 kDa subunits. EcoRI functions optimally at 37°C in its specific buffer and is widely used in DNA cloning, restriction mapping, and other genetic engineering techniques. The enzyme's name follows a convention derived from its bacterial source (E. coli), the strain (RY13), and its status as the first enzyme isolated from this strain. Discovered in 1970, EcoRI was one of the first restriction enzymes isolated and has played a significant role in the development of recombinant DNA technology. It is sensitive to CpG methylation but not to Dam or Dcm methylation. EcoRI's ability to create specific, predictable DNA fragments with sticky ends makes it an invaluable tool in genetic engineering and molecular biology research.
