Rising pH (Reference AOCS Ba 9‐58) Apparatus: Water Bath, pH Meter, Test Tubes with stopper (20mm x 150mm) Reagent: 1.Phospahte buffer solution (0.05M)‐ Dissolve 3.403gm of reagent grade monobasic potassium phosphate (KH2PO4) in approximately 100ml of freshly distilled water. Dissolve 4.355gm of reagent grade dibasic potassium phosphate (K2HPO4) in approximately 100 ml water. Combine the two solution and make exactly 1000ml. if reagent is pure pH should be at 7.0. if it is not. Adjust to 7.0 with a solution of an acid or base before using. The useful life of the buffer solution, prepared us described, is less than 90 days. 2. buffer urea solution: ‐ dissolve 15gm urea reagent grade in 500ml of the phosphate buffer solution. Add 5ml of toluene to serve as a preservative and to prevent mold formation. Adjust the pH of the urea solution to 7.0 as in reagent 1. Preparation of sample: 1. Grind the sample finely as possible with mixing the temperature and mix completely. At least 60% of the sample should pass a no. 40 us standard sieve. Procedure: 1. Weigh 0.2gm (+‐0.001 g) of sample into a test tube and add 10ml of the buffered urea Solution. Stopper mix and place in water bath at 30C. do not invert the tube during the process of mixing. 2. Prepare a blank by weighing 0.2gm sample into a tube 10ml phosphate buffer solution. Stopper, mix & place in water bath at 30C. allow a time interval of 5mint between the preparation of the test and the blank portions. Agitate the content of each tube at 5mint interval. 3. Remove the test tube and blank from the water bath after 30 mints. Transfer the supernatant liquid another tube, maintaining the 5mint interval between the test and blanks. Determine the pH of the supernatant liquids at exactly 5 mint after removal from the both.
