This video provides a comprehensive animated overview of the Transcription-Mediated Amplification (TMA) and Real-Time TMA (RT-TMA) processes that underlie the detection and quantification of pathogens on the Panther® system. TMA is an isothermal nucleic acid amplification technology that can be used to amplify RNA and DNA sequences. The entire reaction takes place in a single tube on the fully automated Panther System. Initially, pathogens are lysed to release nucleic acid molecules. During the target capture step, capture oligonucleotides hybridize to target genetic material of the pathogen. Isolated specimens are separated using magnets. In the amplification phase, reverse transcriptase synthesizes a DNA copy of the target nucleic acid. The DNA template is then transcribed by T7-RNA Polymerase to produce multiple RNA amplicons, which are then copied to produce more cDNA molecules, creating a cycle of amplification. Our qualitative detection with hybridization methods uses chemiluminescent probes to confirm the presence of pathogen genetic material which increases the signal through amplification over time.
