Imaging plate readers, are exactly that - plate readers that have been adapted to take images, usually with a CCD camera and a second mode on the instrument. Most often, the primary purpose of an imaging plate reader is to read fluorescence or luminescence as a single output feature and determine and measure any kind of on- or off- signal. So, when you add a camera and a different light path, that plate reader can become a pseudo high-content analysis machine. HCA platforms focus on high-resolution imaging and the parallel analysis that goes along with the imaging. This assay-centric approach to understand cell-level phenotypes is more closely related to a high-end automated microscope rather than a lower-resolution plate reader. There are 2 main areas where imaging plate readers differ. High-content analysis platforms are designed to adapt to both assay sensitivity and biological sensitivity at the cell level. With a primitive optical path and less-sensitive camera, imaging plate reader assays are less sophisticated and may be limited due to algorithm or system limitations. Secondly, multimode plate readers do not have advanced system features such as laser autofocus, confocal modes, and various other tools related to a true microscope.
