This video describes the process of mounting a slide, viewing and measuring spores using a reticle. This video only goes up to 400x total magnification, but for the most accurate measurements, you want to use the 100x oil immersion objective, for a total of 1000x magnification. This involves adding a drop of immersion oil to the top of the slide before going to the highest magnification.
