Polyploidization is the multiplication of the whole chromosome complement and has occurred frequently in vascular plants. Maintenance of stable polyploid state over generations requires special mechanisms to control pairing and distribution of more than two homologous chromosomes during meiosis. Why Investigate Polyploidization? Polyploidization is considered as a valuable tool in the genetic improvement of crop plants. The evolutionary success of polyploid plants in nature and under domestication has been attributed to buffering of mutations. Leaf and Stomata Morphology of Mature Fan Leaves Leaves collected after 4 weeks of vegetative growth and 1 week under flowering lights Nail polish impressions show stomata on the abaxial surface of fan leaves Leaves have very similar morphology Stomata appear larger in size on the tetrapoloid plants Trichome Density Images show trichome density on the adaxial surface of the 4th sugar leaf of C. sativa strain Plants A and B = Diploid Plants C and D = Tetraploid Leaves were imaged on the 7th week of flowering Note: Scale bars, 1mm Inflorescence Architecture Inflorescence apex (cola) and buds shown for two plants during the 8th week of flowering of C. sativa strain. Week 12 after transplanting and 1 week before harvest Plant A and C = Diploid Plant B and D = Tetraploid Images A and B represent Cola Images C and D represent bud morphology Phytochemical Content Dried Buds and leaves of C. sativa plants of different ploidy were assessed by HPLC and GC for cannabinoids and terpens A = Cannabinoid Profile B = Terpen Profile Data are means +/- standard error with 10 diploids and 9 tetraploids analyzed Means with different upper/lower case letters are significantly different ANOVA with Tukey’s post hoc test, p greater than 0.05 Summary Phytochemical content of lower in leaves is high enough for the trimmed leaf material to be used for extraction in both ploidy levels studied which could almost double total production yield. Total terpene content of tetraploid leaves is comparable to the diploid bud. Terpene extracts from tetraploid leaves may have commercial value despite low levels of cannabinoids.
