Interventional Immunotherapy: Transforming the Treatment of Solid Tumors

“Interventional Immunotherapy: Transforming the Treatment of Solid Tumors” was presented at the Society of Interventional Oncology (SIO) 2022 Annual Scientific Meeting. This video describes two key barriers associated with immunotherapy treatment of solid tumors in patients with primary or metastatic liver cancers and also offers information about the Pressure-Enabled Regional Immuno-Oncology™ (PERIO™)-01 clinical trial, sponsored by TriSalus Life Sciences®. In this video, Dr. Alexander Kim, MD, who currently serves as the medical director at National Vascular Physicians and director of interventional oncology clinical strategies at TriSalus Life Sciences, describes the impact of high intratumoral pressure (ITP). ITP impacts effective delivery of immunotherapy treatment to tumors. This is caused by two factors: interstitial fluid pressure, which limits the ability of the therapy to leave blood vessels and deeply penetrate the tumor tissue, and solid stress, which collapses blood vessels and prevents drugs from reaching malignant cells downstream. Dr. Steven C. Katz, MD, FACS, the Chief Medical Officer at TriSalus Life Sciences, then provides information on the role of a second barrier called immunosuppression. As cancer cells alter the environment in the liver, they can further suppress the immune system and decrease the efficacy of immunotherapies. Dr. Rahul Sheth, MD, who serves as an associate professor of interventional radiology at MD Anderson Cancer Center, then provides an overview of the PERIO-01 clinical trial. This trial is studying a new investigational drug, SD-101, delivered intravascularly by the TriNav® Infusion System using the Pressure-Enabled Drug Delivery™ (PEDD™) method of administration. The study is evaluating whether this platform approach can improve the performance of systemic checkpoint inhibitors in treating uveal melanoma with liver metastases by overcoming two major challenges of treatment: immune response suppression and ineffective drug delivery.
