FACS OFF! Autonomous & High Throughput Single Cell Cloning B-cell cloning is an industry standard method for reliable and reproducible production of monoclonal antibodies (mAb). A typical journey in mAb development involves B cells isolation prior to screening. For years, FACS has been the method of choice for semi-automated seeding of single cells into microwell plates. However, this method suffers from low throughput due to inherent difficulties to automate start up, calibration, sample/target exchange processes. Furthermore, FACS is labour-intensive as it requires continuous presence of skilled operator to monitor and troubleshoot the process. Hence, the need of an alternative approach that addresses the aforementioned shortcomings that are: throughput, clonability, and high operating costs. In this webinar, we will present how cellenONE HT, an image-based single cell isolation platform, is fully automated using Green Button Go Scheduler so that it operates continuously without the need for an operator, effectively enabling 24/7 unattended single cell plating from one or multiple samples. Frequently Asked Questions (FAQ) Question 1: Can the system tell the difference between live and dead cells? Yes, the cellenONE HT is equipped with fluorescence, so you can have a usual red/green marker, which allows you to sort. So if you want to sort only live cells, you can do that. Question 2: Does cellenONE HT work with samples in 96 well plates? Yes, samples can be placed in 96 well plates. Question 3 : Why is the clonal outgrowth better with the cellenONE HT? This is thanks to our proprietary dispensing technology based on a very gentle piezo-acoustic stimuli to generate the drops. Question 4: What is the minimum sample volume for a campaign? The dead volume of the sample container is less than 2 µL and we recommend a cell density of around 150 cells/µL. For isolating cells in 4 x 384 well plates we recommend a volume of around 15 µL. Question 5: Can cellenONE HT be operated without Green Button Go (Scheduler)? Yes, the cellenONE HT comes with its own proprietary software that can be used to operate our technology as a standalone device. Question 6: What is the volume and variation of the drop?
