Welcome back to Immunology Insights: Basics and Beyond. In our last chapter, we explored next-generation checkpoint therapies and how combination immunotherapy tries to outsmart tumors by targeting several inhibitory pathways at once. Today, we stay in the world of cancer immunotherapy—but shift from drugs that modulate immune responses to therapies that physically add immune cells back into the patient. We’ve already discussed CAR T cells as one of the great success stories in hematologic malignancies. But CAR Ts are only one branch of a broader family called adoptive cell transfer (ACT). In this chapter, we’ll look beyond CAR T cells and explore three other important ACT strategies: TCR-engineered T cells, Tumor-infiltrating lymphocyte (TIL) therapy and Natural killer (NK) cell therapies. Each of these approaches comes with its own strengths, limitations, and clinical niches. So—let’s talk science!
