Towards a mechanistic understanding of genome regulation Speaker: Patrick Cramer, Max Planck Institute, Germany Chair: David Rueda, Imperial College, London Co-Chair: Stephen McLaughlin, MRC Laboratory of Molecular Biology, UK Patrick Cramer briefly mentions how his group solved the structure of the Corona Virus polymerase during the lockdown in Germany and how this has given an insight into drug action. In the main section, he described how structural information has enabled an understanding of the mechanism and regulation of gene transcription, through remodelling nucleosome position by RSC, binding of pre-initiation complexes, opening the DNA, polymerase recruitment, early polymerase pausing, polymerase release and activation followed by passage through nucleosomes and co-transcriptional pre-spliceosome assembly.
