Fab-TACS® is based on the IBA’s universal Strep-tag® technology and allows label-free cell isolation from different samples via flow cytometric sorting, magnetic separation or affinity chromatography. In all methods, low-affinity Fab fragments bind specifically, but only weakly, to the target markers on the cells’ surface. The Fab fragments are fused to a short peptide sequence called Twin-Strep-tag® (= Fab-Streps). The tag is used to multimerize the Fab fragments on a streptavidin variant called Strep-Tactin. This multimerization increases the binding strength sufficiently to allow efficient capture of target cells. Biotin addition disrupts the multimerization by causing the release of Fab-Streps from Strep-Tactin®. The monomerized Fab-Streps spontaneously dissociate from the cell surface due to their low affinity, which results in a completely label-free cell population. Cells are only minimally affected during the isolation procedures and suitable for various downstream applications. This video tutorial demonstrates all steps involved in cell isolation via affinity chromatography using Strep-Tactin® TACS Agarose columns, including reagent, sample and column preparation.
