Start by completing main story progress by taking out the main target; The Mourner. After this, you'll be able to find Nobutsuna near Azuchi, available on the People of Omi objective board. Talk to them to start the Nobutsuna's Student side quest, which involves taking out 6 students in duels around the open world. The quest culminates in a fight against Sanada Masatoyo and unlocks the Samurai Stand ability in Yasake's Samurai skill tree. Don't forget to assign the skill as well.
