Medical Physics Masters presentation by: Allison Ng Title: Optimising combination cancer therapies: searching the massive solution space of a mechanistic model Aims: This project aims to explore the optimal combination of radiotherapy (RT) and immune checkpoint inhibitors (ICI) such as anti-PD-L1 and anti-CTLA-4 for treating mesothelioma. Immunotherapy (IO) primes the immune system to target cancer cells, with ICIs blocking proteins that enable cancer cells to evade immune detection. Although ICIs have improved patient outcomes, their efficacy is limited, with response rates of 15-30% in solid tumors and adverse side effects in some patients. Research at UWA focuses on enhancing the response to IO by modifying the tumor environment through RT, leveraging pre-clinical data showing synergistic effects of RT and IO in other cancers. The project will use computational models on murine data to identify treatment combinations that maximize tumor treatment probability while being robust to uncertainties.
