He eventually became a bandleader in his own right, and formed his own orchestra in the 1950s. His music was a fusion of traditional Cuban rhythms such as rumba, mambo, and cha-cha-cha, with jazz and other influences. He was known for his skill as a bandleader and arranger, and his orchestra included some of the top musicians of the day. Barreto's music was popular in both Cuba and the United States, and he recorded a number of albums for various record labels. He also performed in nightclubs and other venues in both countries, and his music was featured in films and television shows. Although he is not as well known today as some of his contemporaries, Don Marino Barreto Jr. and His Cuban Orchestra made a significant contribution to the development of Cuban music and its fusion with jazz and other genres.
