Even though the blocking reagent present on the membrane should ideally prevent any primary antibody from binding directly to the membrane, we cannot guarantee that every single open spot on the membrane is covered with the blocking reagent. Therefore, the washing steps are critical because they will remove the excess of unbound antibody and of non-specifically bound antibody.
