The route runs from Kōchi City, Kōchi Prefecture, northward along the Niyodogawa River, Kamiyakawagawa River, Edagawagawa River, Ōmorigawa River, Yoshinogawa River, Kuwazegawa River, Tanigawa River, and Kamogawa River to Saijō City, Ehime Prefecture, and is known as the shortest route between central Kōchi Prefecture and Tōyo region, Ehime Prefecture. The 11.9 km section from the Kōchi Prefectural Office at the starting point to the east end of the Niyodogawa Bridge in Inochō, Agawa District, overlaps with Route 33, and the 5.8 km section from Shimoyakawa-kō to Kamiyakawa-kō in Inochō, Agawa District, overlaps with Route 439.
