RNasin is a ribonuclease (RNase) inhibitor originally derived from human placenta, known for its ability to protect RNA from degradation by binding to RNase enzymes. This protein, with a molecular weight of approximately 51,000 Da and an isoelectric point of 4.7, forms non-covalent complexes with RNase, effectively inactivating it. RNasin is active in buffer conditions of 0.5 M NaCl, pH 5-8, with optimal activity at pH 7.8. It specifically inhibits RNase A, B, and C in eukaryotes, as well as human placental RNase, but does not affect RNase H, S1 nuclease, various RNA polymerases, reverse transcriptases, or DNA polymerases. This specificity makes RNasin invaluable in molecular biology applications where RNase contamination could compromise RNA integrity, such as during cDNA synthesis, in vitro transcription and translation systems, and the preparation of RNase-free protein products like antibodies. Typically, 0.5-1 μl (about 20-40 units) of RNasin is added to a 20 μl reverse transcription system to ensure RNA protection. One unit of RNasin is defined as the amount required to inhibit 50% activity of 5 ng RNase A. By safeguarding RNA, RNasin is an essential tool for researchers working with RNA in various experimental settings.
