A small amount of the food extract is placed in a test tube, followed by the addition of Benedict's solution. The mixture is then heated in a water bath for about five minutes. If glucose or another reducing sugar is present, the solution changes color from blue to green, yellow, orange, and finally forms a brick-red precipitate. Sucrose Test: Sucrose, being a non-reducing sugar, requires hydrolysis before testing. A sample of the food extract is boiled with a few drops of dilute hydrochloric acid to break sucrose into glucose and fructose. The solution is then neutralized using sodium hydroxide. The Benedict's test is performed afterward. A brick-red precipitate after this process indicates the presence of sucrose. Starch test: To test for starch, iodine solution is used. A small amount of the food extract is mixed with a few drops of iodine solution. If starch is present, the mixture turns blue-black. This is a simple and quick test to detect starch in food samples. Fat or Oil Test: In the grease spot test, the food sample is rubbed onto a piece of filter paper. If fat is present, the paper becomes translucent when held up to the light. Protein Test: The Biuret test is used to detect proteins. A small quantity of the food extract is treated with sodium hydroxide solution, followed by a few drops of copper sulfate solution. If proteins are present, the mixture changes to a violet or purple color. This color change confirms the presence of peptide bonds in the protein molecules.
