Xiuning Le, MD PhD, The University of Texas MD Anderson Cancer Centre, Houston, TX, outlines the outcomes of a Phase II study (NCT03318939) of poziotinib, a tyrosine kinase inhibitor (TKI), in HER2 exon 20 insertion non-small cell lung cancer (NSCLC). The study evaluated the efficacy of poziotinib in patients who progressed after chemotherapy and trastuzumab deruxtecan, an FDA-approved antibody-drug conjugate (ADC) for patients with HER2+ NSCLC. The trial enrolled eligible patients from the ZENITH20 trial (NCT03318939) who had received two lines of therapy. Results demonstrated a preserved progression-free survival, tumour response and safety. Dr Le suggests the report will be useful in strategizing for patients who progressed after chemotherapy, and the study confirms the hypothesis that small molecular therapy preserves the same efficacy after ADC therapy.
