The undisputed "mother" of all #cancer mutations: #TP53. Small, nasty, you don't want it - but why not? TP53 has been known as a #tumor suppressor gene since 1979, even before oncogenes. Much like KRAS, it's always had a bad rap (when mutated), especially since we're talking about it here v. A. Have to do with inactivating mutations - the (vast majority of) mutations involve a loss of function of the protein, which eventually can no longer perform its core function. As a result, the tumor accumulates mutations. It can - but doesn't have to! - Have an impact on therapy... TP53 is mutated in up to 50% of all cancer diagnoses without leading directly to cancer. In addition to all types of possible acquired mutations, a hereditary form also plays a major role in Li-Fraumeni syndrome. Here the inherited TP53 mutation does not cause cancer per se, but sooner (infancy) or later (young adulthood) the genetic instability means that a driver mutation can "take hold" and thus lead to cancer.
