In this video, I introduce the membrane curvature evolution that occurs during clathrin-mediated endocytosis (CME), and I discuss the clathrin lattice’s ubiquity on plasma membranes of different shapes. I explain the nanofabrication method we employ to de-couple curvature generation from clathrin-mediated endocytosis. I show that curvature localizes the endocytic machinery, and demonstrate the enrichment of endocytic proteins on curvature as an important measure of CME’s fidelity; I then show that curvature drives the nucleation of endocytic sites in the context of FCHo1/2 knockdown. I then move to clathrin knockdown, in which I show that endocytic localization to nanoscale curvature is independent of the clathrin coat; I also show that sites localized to curvature bear the canonical fluorescence traces of CME, despite the disruption of the clathrin coat. I show through confocal imaging of fluorescent cargoes that curvature rescues the uptake of endocytic vesicles after clathrin knockdown. These data show that CME preferentially nucleates on membranes that have curvature, and that clathrin’s essential function during CME is to stabilize the curvature of the evolving vesicle.
