What You’ll Need: Pipette (e.g., 20-200 μL model) 0.1 mg Balance Empty Beaker Distilled Water Pipette Tips Calibration Tools Step 1: Room Preparation Place the pipette and distilled water in a temperature-controlled room (20 ± 5°C, 40%-70% humidity) for at least 4 hours before calibration. Ensure there’s no ventilation during calibration. Step 2: Set Pipette to Minimum Volume Adjust the pipette to 20 μL by turning the volume adjustment knob counterclockwise. Press the lock to secure the setting. Step 3: Prepare Balance Ensure the balance is calibrated and tared to zero. Step 4: Aspiration and Dispensing Aspirate 20 μL of distilled water and dispense it into the empty beaker. Wait for the balance reading to stabilize. The expected weight should be 0.020 g. If the reading is higher, calibration adjustments are needed. Step 5: Calibration Adjustment Insert the calibration tool, align it with the pipette’s opening, and press down while rotating the volume adjustment lock. Adjust clockwise if the weight is too high or counterclockwise if it’s too low. Repeat until the reading is accurate.
