PUMA is a DIY open source portable 3D printed microscope with augmented reality, fluorescence, polarisation, dark ground, phase contrast, epi-illumination and other advanced features. Beam splitters are essential for more advanced light microscopy techniques such as co-axial epi-illumination and binocular heads. For PUMA a splitter is also used as a beam joiner for the Augmented Reality Heads Up Display (AR HUD). This video includes a tutorial section about beam splitters and also illustrates several RMS / DIN standard lengths and distances used in microscopy such as the mechanical tube length and par-focal distance, etc. At the end I show you how to build the two beam splitter components used in PUMA - the advanced filter block and the splitter component of the binocular head.
