In this video I have explained What is Positive selection and What is Negative Selection I have also explained the difference between positive negative Selection. Positive selection of Thymocytes take place in cortex region of Thymus while the Negative Selection of T cells take place in cortex as well as Medulla of Thymus. Positive selection is also called MHC Restriction while Negative Selection is called Central Tolerance I have explained MHC Restriction in Hindi and Urdu In next Video I will explained Central Tolerance in Hindi and Urdu Cortical Thymic epithelial cells (cTEC) express the MHC-I and MHC-II if DP Thymocytes identify Self MHC these cells are selected to become Single Positive Thymocytes Positive Selection is based on strength of Affinity between self MHC and TCR of DP thymocytes If a weak or no interaction take place between TCR and self/MHC molecules then these DP thymocytes are lead to death and this step is called Death by Neglect If a moderate interaction take place between MHC and DP TCR then these cells are positively selected and are being converted from DP to SP thymocytes. If a very strong Interaction take place between TCR and self MHC then these DP thymocytes are negatively selected.
