Immunotherapy has become a field of growing interest in the fight against cancer. Once activated by anti-CD3 and anti-CD28 co-stimulation, T cells proliferate and initiate a cell-mediated immune response that is able to kill tumor cells. Historically two-dimensional (2D) monolayer culture models have been commonly used in cell-based assays. However, 2D has certain limitations such as modeling cell-cell and cell-extracellular matrix interactions. In contrast, 3D culture models accurately mimic various features of in vivo tumors, such as spatial architecture, physiological responses, secretion of soluble mediators, gene expression patterns, and drug resistance mechanisms. With the Agilent xCELLigence RTCA eSight 3D Tumor Spheroid Immune Cell Killing assays, you can monitor and quantify killing efficacy of 3D single tumor spheroids by live cell imaging using a combination of brightfield plus 3 channel fluorescence (red, green, and blue) – all inside your incubator. Easily calculate spheroid phenotypic changes (i.e. growth or shrinkage) over time. The eSight generates highly reproducible data, and derives deeper and more meaningful biological insights; while the suite of software tools enables automated calculation of metrics such as fluorescent intensity, diameter, and area.
