In this video, I discuss the differences between Competitive and Noncompetitive Inhibitors and how they affect Km, Vmax, and the Linweaver-Burk (Double Reciprocal) Plot. Competitive inhibitors “compete” with the substrate for binding at the active site. Thus, the competitive inhibitor blocks the substrate from binding, and the substrate and inhibitor cannot both be bound to the enzyme at the same time. It follows that, because of this, competitive inhibitors lower the enzyme’s affinity for its substrate, thus increasing the Km, changing the x intercept. Vmax, however, can still be reached by adding enough substrate to “out-compete” competitive inhibitor for binding enzyme active sites. So, Vmax remains the same, as does the y intercept. Noncompetitive inhibitors bind away from the active site. Thus, the enzyme can bind both the substrate and the inhibitor at the same time. However, this enzyme-substrate-inhibitor complex will not effectively proceed towards products at the normal rate. So, Vmax is decreased. Km, however, remains the same because the substrate can bind the enzyme whether or not the inhibitor is bound to the enzyme; so, affinity is not altered. Thus, the y intercept is changed, but the x intercept remains the same. The graph in the video depicts precisely how the intercepts change in each case.
