DNA replication in Hindi - This lecture explains about the DNA replication process in Hindi. Watch this video lecture to know DNA replication mechanism in Hindi. DNA replication elongation 5. DNA replication termination E coli is used as a model organism to demonstrate the DNA replication in prokaryotes in Hindi. DNA replication process The synthesis of DNA molecule can be divided into 3 stages: Initiation, elongation, termination DNA replication Initiation E.coli replication origin called oriC consists about 245 bp in which 3 repeats of 13 bp sequence and 4 repeats of a 9 bp sequence. The total 8 proteins are involved in replication initiation. They open the DNA helix at the origin and established a pre-priming complex. 1) A complex about 20 DnaA protein molecule binds to the 4-9 base pairs repeat in the oriC. 2) With help of ATP and bacterial histone like proteins named HU proteins, DnaA molecules. Successively denature the DNA. In the region of 3-13bp repeats which are A-T rich region. 3) DNA at this time called open complex. Now DnaB protein or helicase binds at the open complex with the help of DnaC and ATP. DnaB is a hexamer clapms. These two molecules then proceeded to unwind the DNA in opposite direction from oriC. This complex is called pre-priming complex. 4) The ssDNA binding protein and DNA gyrase or topoisomerase II are added which stabilizing the single strand and relieves topological stress respectively created by DnaB. DNA replication Elongation 1)Leading strand synthesis:It begins with the synthesis of short I logo nucleotide RNA primer (10-60 bp) by primase at replication origin. Deoxyribonucleotides are then added to this 3' free OH site of primer by DNA polymerase III. This strand synthesis proceeds continuously at the same direction of fork movement. 2)Lagging strand synthesis: This must be accomplished in short fragments (1000-2000 bp), all okazaki fragments, synthesized to the direction opposite to the fork movement. Lagging strand synthesis is more complicated. The overall direction of growth must be 3'-5', but DNA polymerase III added dNTP only in the 5'-3' direction. For the synthesis of lagging synthesis, RNA primer is required, synthesized BT primase. DNA replication Termination E.coli DNA contain a specific site called termination site where two replication forks meet. This termination site contains many proteins, TUS protein coded by tus gene, and an inhibitor DnaB helicase, are responsible for preventing helicase activity of fork. Finally E.coli chromosome produce two interlocking daughter DNA molecule after completion of replication which separated each other with the help of type II topoisomerase and then segregated into two daughter cell.
