KOMET-008: a Phase I trial of ziftomenib combinations in KMT2A-rearranged or NPM1-mutant AML

Aaron Goldberg, MD, PhD, Memorial Sloan-Kettering Cancer Center, New York, NY, discusses the study design and methods of KOMET-008 (NCT05735184), an open-label, dose escalation and expansion study which will assess the safety and tolerability of ziftomenib in combination with either chemotherapy (FLAG-Ida or low-dose cytarabine) or gilteritinib in patients with KMT2A-rearranged or NPM1-mutant acute myeloid leukemia (AML) with or without concurrent FLT3 mutations. Dr Goldberg describes the mechanism of action of ziftomenib, a menin inhibitor, and shares his excitement at the potential of developing a combination therapy which will deepen the responses previously observed with menin inhibitor monotherapy.
