Antibiotics are known to disrupt the gut microbiome, but emerging evidence shows they can also shape immune responses far beyond the gut. In this session, Dr. Kelly McNagny will present research exploring how low-dose antibiotic exposure alters microbial metabolite production—particularly short-chain fatty acids (SCFAs) and bile acid derivatives—with lasting consequences for lung immunity. His team has shown that vancomycin depletes SCFA-producing bacteria, priming innate and adaptive immune cells—including ILC2s, IgE-producing B cells, mast cells, and basophils—and enhancing susceptibility to allergic asthma. In contrast, streptomycin alters bile acid metabolism, enhancing Th1/Th17-driven hypersensitivity pneumonitis via increased lung ILC3 and antigen-presenting cell activity. Supplementation with specific microbial metabolites such as SCFAs and isolithocholic acid reverses these effects, highlighting the microbiome’s role in shaping immune responses with implications for glycan-mediated immune regulation in chronic inflammatory disease.
