Asking you about your least favorite professor gives them insight into how you deal with people you don't really like. And no, you can't say that you got along with all of your professors really well, that's not realistic. This question is the equivalent to an employer asking about your least favorite boss. The key is to be fair about the relationship and determine if there are other things you could have done differently to change the situation. So if the professor was difficult to understand, was there something you could have done to learn more? Could you talk to students who took the class last year? Could you have followed the book more? Acknowledge that a relationship is a two-way street, and while you may not (nor ever) see eye to eye with your professor, the best way to answer this question is to talk about how you did try to make it work, and take responsibility for what you could have tried to do better.
