0:00 Running PCR 2:16 Running restriction digestion 3:12 Two ways of loading and running gels 4:48 Visualizing forward and reverse primer sites 7:39 Measuring digested fragment sizes/sequences on gels This tutorial shows how the Case It simulation performs PCR using controls from the Cystic Fibrosis case as an example. A feature is demonstrated that identifies forward and reverse primer sites along with an animation of the first step of the PCR process, for any DNA sequence. The resulting products are digested by a restriction enzyme and run on a gel (see parts 1 and 2 of the DNA Electrophoresis tutorial for a more complete explanation of features related to that laboratory technique). This free simulation software, available for both PC and Mac at https://www.caseitproject.org/, enables users to perform a variety of laboratory procedures including qPCR, SNP and expression microarrays, restriction enzyme digestion, gel electrophoresis, PCR, ELISA, Southern and Western blots, and dot blots (https://www.caseitproject.org/cases-a.... Case It won a Science Prize from the American Association of Science (AAAS) for its effectiveness in undergraduate biology and high school education. An essay describing the project was published in Science magazine (“Engaging Students in Molecular Biology via Case-Based Learning“ - https://www.science.org/doi/10.1126/s... ). Case It software was developed with funding from the National Science Foundation, which also supported extensive classroom testing and evaluation of project materials.
