Uncouplers of Oxidative Phosphorylation There are certain compounds that can uncouple tor delink the electron transport from oxidative phosphorylation. Such compounds, known as uncouplers, increase the permeability of inner mitochondrial membrane to protons (H). The result is that ATP synthesis does not occur. ETC is not coupled to the ATP formation, so the energy is dissipated as heat. Un-couplers of Oxidative Phosphorylation 2, 4-Dinitrophenol (DNP): It transfers protons across the mitochondrial membrane thereby diverting its flow from F0F1 ATPase. Valinomycin: It transfers K ions, disturbing the osmotic pressure. Gramicidin: It transfers Na+ ions, across the membrane. All the above three are known as ‘ionophores’ i.e. those which disrupt the membrane permeability to ions, thereby uncoupling phosphorylation with ETC. Inhibitors of Oxidative Phosphorylation A. Oligomycin A: It inhibits ATP Synthase (F0F1ATPase). This antibiotic prevents the mitochondrial oxidation as well as phosphorylation. It binds with enzyme ATP synthase and blocks its proton (H+) channels (Fo subunit), which is necessary for oxidative phosphorylation of ADP to ATP. It thus prevents the translocation (re-entry) of protons into the mitochondrial matrix. Due to this, protons get accumulated at higher concentration in the intermembrane space. Electron transport (respiration) ultimately stops, since protons cannot be pumped out against steep proton gradients. B. Atractyloside: Inhibitors of the ADP-ATP Translocase/ Adenine nucleotide transport protein This is a plant toxin. ADP/ATP translocase, which is located on the inner mitochondrial membrane, which transport ATP in exchange of ADP. It binds and inhibits adenine nucleotide transport protein and, thus, blocks the adequate supply of ADP, thereby preventing phosphorylation. As a result, the exchange of ADP and ATP is no longer carried out and the cell dies due to lack of energy.
